The draw for the 2016 Open Sud de France in Montpellier was released on Saturday and Marcos, seeded eighth, opens against Slovakia's Lukas Lacko.
It will be his fourth head-to-head encounter against Lacko, who is at World No. 111 in the Emirates ATP Rankings. Marcos most recently prevailed in three sets in Washington 2013 and he will look to hand the Slovak his 12th defeat in his last 13 ATP level matches.
Marcos, who was the top seed in 2006 when the tournament was held in nearby Lyon, could potentially face this year's No. 1 seed Richard Gasquet in the quarterfinals. Other threats in his half of the draw include Ernests Gulbis, Gilles Simon and Joao Sousa.
